Areas Of Expertise

Remodelling of connective tissue matrices. Chris initially established the location and proliferative behaviour of cells in the periodontal ligament and showed that subpopulations of these cells can undergo self-renewal, a characteristic of stem cells. This work is widely quoted in the connective tissue cell literature. He used actin isoform analyses to establish that a-smooth muscle actin is a differentiation marker for contractile fibroblasts from the periodontium and is required for collagen contraction. Further, he has shown that the actin binding protein filamin A is required for protection of cells in mechanically active environments. Finally, Chris has established an in vitro model for collagen phagocytosis and has shown the mechanisms by which phagocytosis is important in the formation of fibrotic lesions.

Electroinjection of antibodies to block cytoskeletal protein function in cell migration

To optimize the method by which antibodies can inhibit the intracellular activity of actin severing proteins such as gelsolin, in vitro studies were performed first to determine the amount of antibody that would complex known amounts of gelsolin. From estimates of cell number and the amount of protein in the lysates we determined the optimal concentration of purified gelsolin for experiments (1 µM; i.e. 83 µg/ml). Gelsolin was incubated with various dilutions of gelsolin antibody (mouse monoclonal anti-gelsolin antibody; 2C4; from Sigma in electroporation buffer). The gelsolin-antibody complexes were removed with protein A sepharose. The residual, free gelsolin was detected by immunoblotting and the optimal concentration of antibody for complete complexing was calculated (100 µg/ml). For electroporation and severing assays an equivalent molar amount of antibody was used.

The requirement for gelsolin in cell migration in the chemotactic assay

To assess directly the requirement for gelsolin in cell migration in the chemotactic assay, the gelsolin antibody or an irrelevant antibody (pan-cytokeratin antibody, clone PCK-26; diluted in electroporation buffer) were electroinjected into cells by electroporation with a Bio-Rad gene pulser, a capacitance extender and sterile cuvettes. An 800-ul aliquot of 2 X 106 cells/ml was placed into an electroporation cuvette (0.4 cm inter-electrode distance) and the cells were electroporated in a buffer containing 5.37 mM KCl, 0.52 mM KH2PO4, 0.64 mM MgCl2, 0.63 mM MgSO4, 85.5 mM NaCl, 5.8 mM NaHCO3, 0.50 mM NaH2PO4, and 12.5 mM HEPES at a field strength of 250 V/cm and capacitance of 500 µF. The cells were collected by centrifugation, suspended in growth medium without serum and 5X104 cells/well were assayed in the Boyden chamber with a chemotactic gradient for various time periods. The filter-attached cells were fixed and stained with DAPI before counting. About 108 antibody molecules per cell can be introduced into viable cells using this method and that electroinjected antibodies can specifically bind and inactivate specific functional proteins.

Measurement of calcium ion concentration in specific cellular compartments

For analysis of calcium in specific cellular compartments, cells were incubated with 4.5 mM mag-fura 2/AM for 140 mins at 37°C to load ER stores. Estimates of ER store calcium were made with a microscope-based, ratio fluorimeter (Photon Technology Inc., London, ON) in single cells. For mitochondrial calcium, Rat2 fibroblasts were loaded with 4.5 mM rhod-2/AM in 0.005% (v/v) Pluronic gel for 30-140 mins at 37°C. The cells were washed twice and incubated in calcium buffer for imaging. The magnitude of fluorescence of Rhod 2-stained samples was observed in an inverted microscope equipped with a CCD camera (Pentamax; Princeton, NJ) and analysed using the Winview software program (Princeton). As ratio imaging cannot be used to overcome problems of dye leakage and photobleaching in time course experiments with rhod-2, single excitation/single emission imaging analyses were conducted with a number of corrective procedures. First, the fluorescence intensity of rhod-2 was measured in small (~4 mm2) sampling grids in the lamellopodia of well-spread cells to avoid the inclusion of nucleoli which stain brightly with rhod 2. Second, background fluorescence and adjustments for time-dependent photo-bleaching were made for each measurement by calculating the bleach-induced rate constant (i.e. time-dependent loss of fluorescence) in untreated cell samples that were not incubated with collagen-coated beads. Third, for verification of the appropriate spatial localization of the rhod-2 staining and to ensure that rhod-2 was not sequestered to lysosomes, in some experiments Rat2 fibroblasts were co-loaded with 100 nM MitoTracker Green and 4.5 mM rhod-2/AM in 0.005% (v/v) Pluronic for 30 minutes. The cells were washed twice and left in calcium buffer before imaging. These loading protocols facilitate measurements of store-specific calcium after stimulation by a variety of ligands including collagen or fibronectin ligands.
